Abstract Information :

The use of sample preparation to provide enhanced performance is reviewed for the analysis of biological fluids. The impact of using differing sample preparation approaches, including supported liquid extraction, protein precipitation and solid phase extraction is investigated. The comprehensive investigation will determine the effect on recovery, matrix effects and matrix removal. It is shown that the use of protein precipitation can provide robust data, however some of the underlying information demonstrates that this approach may not be suitable for a wide range of compounds. Data will be presented which demonstrates the effect of changing the sample preparation technique on the retention of phospholipids and the impact that this can have the detection robustness and how altering the chromatographic or sample preparation conditions can ensure that the method developed is robust. A series of experiments will be performed that also determine the recovery, and matrix effects for a series of compounds using an approach supported by the regulatory bodies. This provides an interesting insight into the challenges that are associated with bioanalysis.
